This is a working overview of REV-ERB, written for readers who want more than a one-paragraph summary but less than a textbook.
Reviewed 2025-12-01. Anything still debated is marked as such rather than presented as settled.
SR9009 is frequently discussed alongside other REV-ERB ligands, including synthetic agonists and natural heme-related molecules. Its selectivity for REV-ERB over related nuclear receptors has been measured in binding and reporter assays, though off-target activity at higher concentrations is possible. The compound is prohibited in sport by the World Anti-Doping Agency, and it is not approved for any medical use in major jurisdictions. Products sold online may be labeled as research chemicals, and their identity and purity are not guaranteed by regulatory review.
SR9009 is a synthetic small molecule that acts on the nuclear receptors REV-ERBα and REV-ERBβ. These receptors are part of the circadian clock machinery and normally repress transcription of certain target genes. In laboratory research, SR9009 is used as a chemical tool to study how REV-ERB activity influences metabolism, inflammation, and daily biological rhythms. The compound is not an approved medicine, and its effects in humans remain largely uncharacterized. It is often described as an investigational agent rather than a therapeutic product.
REV-ERB proteins typically suppress gene expression by recruiting corepressor complexes to DNA response elements. SR9009 binds these receptors and strengthens that repressive action in cell-based assays. Studies in rodents have reported changes in lipid handling, mitochondrial function, and exercise capacity after treatment. Such findings are often cited as evidence for metabolic effects, but species differences and limited pharmacokinetic data make direct translation to humans uncertain. Researchers continue to examine which effects are robust and which depend on specific experimental conditions.
SR9009 is generally described as poorly soluble in water and more soluble in organic solvents such as dimethyl sulfoxide and ethanol. Stock solutions are commonly prepared in an organic solvent before dilution into an aqueous buffer or vehicle. Precipitation can occur if the organic fraction is reduced too quickly or if the final concentration exceeds the compound's solubility limit. Sonication or gentle warming may aid dissolution in some protocols, but excessive heat can promote degradation. Container material and pH can also influence observed solubility.
For long-term storage, SR9009 is typically kept as a solid at low temperature, protected from moisture and light. Desiccated conditions limit hydrolysis, while opaque containers reduce photochemical breakdown. Solutions are less stable than solids and are often stored frozen in aliquots to avoid repeated freeze-thaw cycles. Stability data are not standardized across all suppliers, so users should rely on certificate-of-analysis information when available. Degradation may appear as color change, precipitate, or decreased chromatographic purity.

Quality control for research materials includes identity confirmation by nuclear magnetic resonance and purity assessment by high-performance liquid chromatography. Mass spectrometry provides molecular weight confirmation and can detect related impurities. Purchasers should request a certificate of analysis that lists lot-specific data. Online products advertised for human use often lack such documentation. Distinguishing legitimate research material from mislabeled or contaminated samples is a recurring challenge in independent testing, and independent laboratories may use orthogonal methods to verify identity.
Detection of SR9009 in biological samples usually relies on liquid chromatography coupled to tandem mass spectrometry. This approach separates the compound from matrix components and identifies it by mass transitions. Because SR9009 can undergo metabolism, laboratories often look for both parent drug and specific metabolites. Sample preparation may involve protein precipitation or solid-phase extraction. Method validation examines sensitivity, carryover, and interference from related substances, and reference standards are required for accurate calibration.
Regulatory agencies have not approved SR9009 for human therapeutic use. It is typically sold as a research chemical with labels stating that it is not for human consumption. The World Anti-Doping Agency prohibits the substance in sport, generally under the category of non-approved substances. Customs and national laws may restrict importation, sale, or possession. Product quality and legal status can vary by country and vendor, and therapeutic claims are not permitted in regulated advertising because the compound lacks approval.
Several misconceptions surround SR9009. It is often described as a SARM, a steroid, or an exercise pill, but its known target is the REV-ERB receptor family. Rodent studies have examined exercise capacity and metabolic markers, yet human outcomes remain unproven. Oral bioavailability appears low in animals, and human pharmacokinetics are not well characterized. Online products may contain impurities or different compounds, so identity and purity testing are important for research use.
It is complex, multi-factorial, and likely to be also represented by a series of sub-groups driven by pain varying from peripheral or central nervous system, altered laxity/stiffness of muscles, laxity to injury of tendinous/ligamentous structures to maladaptive body mechanics.
The antiporter transports the ADP into the matrix, while transporting ATP out. Due to the high concentration of ATP around the mitochondrial creatine kinase, it will convert ATP into PCr which will then move back out into the cells cytoplasm to be converted into ATP (by cytoplasmic creatine kinase) to be used as energy for muscle contraction. In some vertebrates, arginine phosphate plays a similar role.
RuBP + O2 → Phosphoglycolate + 3-phosphoglycerate + 2 H+ During the catalysis by RuBisCO, an 'activated' intermediate is formed (an enediol intermediate) in the RuBisCO active site. This intermediate is able to react with either CO2 or O2. It has been demonstrated that the specific shape of the RuBisCO active site acts to encourage reactions with CO2. Although there is a significant "failure" rate (~25% of reactions are oxygenation rather than carboxylation), this represents significant favouring of CO2, when the relative abundance of the two gases is taken into account: in the current atmosphere, O2 is approximately 500 times more abundant, and in solution O2 is 25 times more abundant than CO2. The ability of RuBisCO to specify between the two gases is known as its selectivity factor (or Srel), and it varies between species, with angiosperms more efficient than other plants, but with little variation among the vascular plants. A suggested explanation of RuBisCO's inability to discriminate completely between CO2 and O2 is that it is an evolutionary relic: The early atmosphere in which primitive plants originated contained very little oxygen, the early evolution of RuBisCO was not influenced by its ability to discriminate between O2 and CO2.
Myxedema's characteristic physical sign is non-pitting edema, in contrast to pitting edema. Myxedema can also occur in the lower leg (pretibial myxedema) and behind the eyes (exophthalmos). Severe cases, requiring hospitalization can exhibit signs of hypothermia, hypoglycemia, hypotension, respiratory depression, and coma.

At the time of the K-Pg extinction, the Deccan Traps flood basalts of India were actively erupting. The eruptions can be separated into three phases around the K-Pg boundary, two prior to the boundary and one after. The second phase, which occurred very close to the boundary, would have extruded 70 to 80% of the volume of these eruptions in intermittent pulses that occurred around 100,000 years apart. Greenhouse gases such as carbon dioxide and sulfur dioxide would have been released by this volcanic activity, resulting in climate change through temperature perturbations of roughly 3 °C (5.4 °F) but possibly as high as 7 °C (13 °F). Like the Chicxulub impact, the eruptions may also have released sulfate aerosols, which would have caused acid rain and global cooling. However, due to large error margins in the dating of the eruptions, the role of the Deccan Traps in the K-Pg extinction remains unclear. Before 2000, arguments that the Deccan Traps eruptions—as opposed to the Chicxulub impact—caused the extinction were usually linked to the view that the extinction was gradual. Prior to the discovery of the Chicxulub crater, the Deccan Traps were used to explain the global iridium layer; even after the crater's discovery, the impact was still thought to only have had a regional, not global, effect on the extinction event. In response, Luis Alvarez rejected volcanic activity as an explanation for the iridium layer and the extinction as a whole.
=== Data analysis === Mass spectrometry data analysis is specific to the type of experiment producing the data. General subdivisions of data are fundamental to understanding any data. Many mass spectrometers work in either negative ion mode or positive ion mode. It is very important to know whether the observed ions are negatively or positively charged. This is often important in determining the neutral mass but it also indicates something about the nature of the molecules. Different types of ion source result in different arrays of fragments produced from the original molecules. An electron ionization source produces many fragments and mostly single-charged (1-) radicals (odd number of electrons), whereas an electrospray source usually produces non-radical quasimolecular ions that are frequently multiply charged. Tandem mass spectrometry purposely produces fragment ions post-source and can drastically change the sort of data achieved by an experiment. Knowledge of the origin of a sample can provide insight into the component molecules of the sample and their fragmentations. A sample from a synthesis/manufacturing process will probably contain impurities chemically related to the target component. A crudely prepared biological sample will probably contain a certain amount of salt, which may form adducts with the analyte molecules in certain analyses. Results can also depend heavily on sample preparation and how it was run/introduced.
The Shihab dynasty was an Arab family whose members served as the paramount tax farmers and local chiefs of Mount Lebanon from the early 18th to mid-19th century, during Ottoman rule. Their reign began in 1697 after the death of the last Ma'nid chief. In 1697, Amir Ahmad died without an heir, and the Druze notables chose his nephew Bashir al-Shihabi as their new ruler. He was succeeded in 1707 by the young Amir Haydar al-Shihabi, grandson of Amir Ahmad al-Ma'ni. Haydar recognized the authority of the Maronite al-Khazins and the Hubayshis of Kisrawan and Ghazir and treated these two families as equal to the feudalistic Druze families. The Shihab family realized the importance of Maronite rule and power, and they and the Maronites became united in a common interest. The family centralized control over Mount Lebanon, destroying the feudal power of the mostly Druze lords and cultivating the Maronite clergy as an alternative power base of the emirate. During Yusuf Shihab's rule, many members of the Shihab family converted to Christianity and Yusuf also began to rely on the support of the Maronite Christians. On 3 September 1840, Bashir Shihab III, a distant cousin of the once-powerful Emir Bashir Shihab II, was appointed emir of Mount Lebanon by Ottoman Sultan Abdulmejid I. Geographically, the Mount Lebanon Emirate corresponded with the central part of present-day Lebanon, which historically has had a Christian and Druze majority. In practice, the terms "Lebanon" and "Mount Lebanon" tended to be used interchangeably by historians until the formal establishment of the Mandate.
=== United States herbalism fraud === Over the years 2017–2021, the US Food and Drug Administration (FDA) issued warning letters to numerous herbalism companies for illegally marketing products under "conditions that cause them to be drugs under section 201(g)(1) of the Act [21 U.S.C. § 321(g)(1)], because they are intended for use in the diagnosis, cure, mitigation, treatment, or prevention of disease and/or intended to affect the structure or any function of the body" when no such evidence existed. During the COVID-19 pandemic, the FDA and US Federal Trade Commission issued warnings to several hundred American companies for promoting false claims that herbal products could prevent or treat COVID-19 disease.
Austin further cemented his heel turn the following Thursday on SmackDown!, when, during an interview with Jim Ross about his actions at WrestleMania, he thought Ross was denouncing their friendship and then assaulted Ross. Austin and Triple H became a team known as The Two-Man Power Trip. Austin altered his character considerably over the next few months by becoming a whiny, temperamental prima donna who complained incessantly when he felt he was not getting respect. He also developed a strange infatuation with McMahon, going to great lengths to impress him, even going so far as to hug him and bring him presents. Austin and Triple H ran roughshod over all their opponents, until coming up against The Undertaker and Kane. After defeating them for the WWF Tag Team Championship at Backlash on April 29, they held the tag team titles, the WWF Championship (Austin) and the Intercontinental Championship (Triple H) all at once. On the May 21 episode of Raw is War, Austin and Triple H defended the WWF Tag Team Championship against Chris Benoit and Chris Jericho; during the match, Triple H legitimately tore his quadriceps, and the team lost the match and the tag team championship. Austin officially broke up The Power Trip on that week's SmackDown!, criticizing Triple H for his injury and for hitting him with the sledgehammer. He continued to align himself with McMahon and began feuding with Jericho and Benoit by himself, leading to a triple-threat match at King of the Ring on June 24; despite interference from the debuting Booker T, Austin retained the championship.

This compound is used as a component in the production of fire-extinguishing compounds, pharmaceuticals, dyes, pigments, and it is also a basic fertilizer, being a source of ammonia. Ammonium bicarbonate is still widely used in the plastics and rubber industry, in the manufacture of ceramics, in chrome leather tanning, and for the synthesis of catalysts. It is also used for buffering solutions to make them slightly alkaline during chemical purification, such as high-performance liquid chromatography. Because it entirely decomposes to volatile compounds, this allows rapid recovery of the compound of interest by freeze-drying. Relatedly it is also useful as an alkaline buffering agent for analytical LC–MS as its volatility allows it to be rapidly removed automatically from the sample stream in the low pressure spray chambers used by many standard mass spectrometry detectors found at the end of typical LC-MS systems, such as electrospray ionization detectors. This is critical as most mass spectrometry detectors become signal saturated or even damaged with more than a trace amount of ions entering the detector proper at any one time. This issue limits buffering agents and other additives in LC-MS buffers to either extremely trace concentrations or to fairly volatile compounds. In pH ranges from about 7 to 9, ammonium bicarbonate is one of the only options available as the primary buffering agent for most LC-MS buffers. Ammonium bicarbonate is also a key component of the expectorant cough syrup "Senega and Ammonia".
The most studied ring destructive pathway is the Kodama pathway and it was initially identified in Pseudomonas abikonensis and Pseudomonas jijani. The pathway comprises four main steps: i) the successive hydroxylation by NADH-dependent dioxygenases of the carbons in one of the aromatic rings, followed by ii) the dehydrogenation of the ring by a NAD+ cofactor and further iii) oxygenation promoting ring cleavage and formation of a pyruvyl branch; concluding with iv) the hydrolysis of the pyruvyl substituent to release pyruvate and the remaining of the substrate. Since the end products of the pathway are still water soluble sulfur compounds, the pathway has often been disregarded as an appealing pathway for industrial applications, in particular by the oil industry. The most well-studied sulfur specific pathway is the 4S pathway, first discovered in the bacterium Rhodococcus erythropolis (strain IGTS8), which was observed to remove sulfur from dibenzothiophenes and derivatives in three steps: i) a double oxidation of the sulfur (to sulfoxide and sulfone) performed by a flavin-dependent monoxygenase, followed by ii) a carbon-sulfur bond cleavage by a second flavin-dependent monoxygenase and a iii) desulfination reaction through which 2-hydroxybiphenyl and sulfite are produced.

== Safety and regulations == PVDF is widely considered safe and ubiquitous used for water treatment, the food industry, and biocompatible devices like hernia meshes or internal devices. PVDF differs from PFAS in that alternating groups are hydrogen, making it less resilient to high temperatures, but also meaning that byproducts don't degrade into known hazardous PFAS. However, studies examining ecotoxity have shown that very high concentrations (up to 100 mg/L) may alter jellyfish behavior, while not being toxic to them. In the US, FDA regulations consider PVDF to be food safe, while US EPA water treatment regulations on PFAS have avoided placing limits on PVDF, while strictly limit concentrations of PFAS. PVDF was added to the Living Building Challenge (LBC) Red List in 2022. The Red List bans substances prevalent in the building industry that pose serious risks to human health and the environment from construction that seeks to meet the criteria of the Living Building Challenge (LBC). Proposed regulations in the EU aim to ban "any substance that contains at least one fully fluorinated methyl (-CF3) or methylene (-CF2-) carbon atom (without any H/Cl/Br/I attached to it)". Unless exemptions are made, the application of inconsistent and severe regulations may propose an existential risk to the industry.

SR9009 is a synthetic research compound that binds and modulates the nuclear receptors REV-ERBα and REV-ERBβ. It is used in laboratory studies of circadian biology and metabolism, not as an approved medicine. It is also known by the informal name Stenabolic.
No. SR9009 has not been approved as a therapeutic drug in the United States, European Union, or other major markets. Human safety and efficacy data are very limited. Its presence in consumer products does not imply regulatory approval.
The World Anti-Doping Agency lists SR9009 as a prohibited substance because it can alter gene expression and metabolic pathways relevant to performance. The ban applies at all times, not only during competition. Detection relies on laboratory methods such as mass spectrometry.
Liquid chromatography with mass spectrometry is a common approach. Ultraviolet detection and nuclear magnetic resonance can support identification when suitable standards are available.
